Lowicryl K11M kit (DG)
Lowicryl resins are highly crosslinked methacrylate embeding media, designed for use over a wide range of embedding conditions. When they are of the correct hardness they are easily sectioned with either glass or diamond knives.
Lowicryl resins are photopolymerised by long wavelength (360nm) ultraviolet light. Since the initiation of the polymerisation is largely independent of temperatures, blocks may be polymerised at the same temperatures used for infiltration. The resin may also be chemically polymerised at 60 deg. C. The K11M kit can be used down to -60 deg. C and is hydrophilic.
The hydrophilic properties of K11M provide two distinct advantages.
- specimens may be kept in partially hydrated state during dehydration and infiltration
- better structural preservation and improved preservation of antigenicity makes K11M particularly useful for immunolabelling of sections
As with other polar (water-miscible) resins, precautions should be taken to insure that the block face does not become wet during sectioning. Sections should be collected as soon as possible after they are cut, and staining incubation should be kept as brief as possible. Optimal sectioning can be done easily with uranyl acetate and Reynold's lead citrate.
